一种鹅病毒病病原的形态观察和理化特性的测定

【摘要】 为更好地了解鹅病毒病病原的形态和理化特性,用鸡胚尿囊液(EVF4)进行试验:用2种红细胞(1种用甲醛醛化,另1种不醛化)进行吸附和纯化,最终得到EVF4-1和EVF4-2,并用电镜观察,进行血凝和血凝抑制试验,毒力及理化特性测定.结果表明:该病毒形态近似球形,有囊膜,直径大小为120~220nm;其血凝效价从高到低依次为鹅、豚鼠、山羊、牛、兔、猪、鸡、鸭、人的“O”型红细胞,对小白鼠红细胞不凝集;血凝抑制试验时,发现能被相应的血清和鸡新城疫血清所抑制;对ELD50为10─5.3,鸡胚平均死亡时间为96h;对乙醚、氯仿敏感;胰蛋白酶不敏感;不耐酸,对热较敏感.

【Abstract】 Goose embryo-adapted virus GVF6 was inoculated into chicken embryo and passaged for many generations.And thus,chicken embryo-adapted virus EVF4 was obtained.Two kinds of virus samples,EVF4-1and EVF4-2,were obtained by two kinds of red blood cells(one fresh,another formaldehyde-treated) adsorption-elution and ammonia sulfate precipitation.The virus was observed under electron microscope for hemagglution test(HA)and hemagglution in- hibition test(HAI) and mcasured in aspect of toxicity,hemaqqucal properties and physiochemical properties.The results showed that virions were enveloped with spherical icosahedriron and 120 nm to 220 nm in diameter.The outcome sequence of Hemagglution test(HA) were geese,guineapigs,sheep,cows,rabbits,pigs,chicken,ducks,human beings’ “O” type red cells from high to low while for mouse no Hemagglution.Positive results indicated that concerning chicken embryos median lethal,if ELD50 was 10─5.3,chicken embryos died after being infected with all the antibiotic fluids in 96 hours.The virus was susceptible to aether,chloroform,and heat.However,it was not sensitive to trypsin and not acid-resistant.
