一株纤维素分解菌的分离及其粗酶性质研究

【摘要】 从堆肥、牛粪、马粪、土壤等样品中共分离出35株微生物菌株,其中的1株霉菌F10 表现出最高酶活力.对比研究发现,F10 在固体发酵实验中具有接近于绿色木霉AS3 3711的酶活力,而在液体发酵实验中则高于绿色木霉AS3 3711.研究结果表明,当碳源物质为羧甲基纤维素(CMC)、氮源物质为硝酸铵时,F10 具有最佳产酶效果,产酶时间为4 8~84h ;当pH为5 0 ,温度在4 0~6 0℃时,相应的滤纸酶活力、CMC酶活力及棉花酶活力最高为16 6、4 3 6、0 4U/g ;而当pH位于4 0~5 0 ,温度为5 0℃时,相应的滤纸酶活力、CMC酶活力及棉花酶活力最高分别为9 1、2 9 5、0 2 5U/g ;当温度超过6 0℃时,酶活力急剧下降.该研究可为生物肥料工业和发酵饲料工业的微生物学研究提供借鉴方法

【Abstract】 Thirty-five strains of cellulose-decomposing microorganisms were isolated from manures of horse, cattle, compost and soil. Among them, F10 had the highest enzyme activity. With AS3.3711 as the enzyme activity reference, F10 demonstrated a quite satisfactory behavior in the related solid fermentation studies; its liquid fermentation results were even a little higher than that of the AS3.3711. Experiments also showed that F10 had the best performance enzyme producing property when carboxymethyl cellulose (CMC) was chosen as the carbon source, ammonium nitrate as the nitrogen source, and the relevant time scale for it was set in 48~84 h. When pH was 5.0 and temperature was from 40 ℃ to 60 ℃, the highest enzyme activities of filter paper, CMC as well as cotton could go up to 16.6, 43.6 and 0.4 U/g, respectively;while temperature was fixed at (50 ℃ and) pH was from 4.5 to 5.0, the correspondent highest enzyme activities were 9.1, 29.5 and 0.25 U/g. When the temperature was higher than 60 ℃, the enzyme activities descended rapidly. This research provided lessons for biology fertilizer industry and fermenting feedstuff industry.
