MTT法测定猪TF对犬淋巴细胞转化增殖的研究

【摘要】 无菌采集犬抗凝血,分离淋巴细胞并制备成不同浓度。应用正交试验,探讨不同浓度淋巴细胞、犊牛血清(FCS)、植物血凝素(PHA-P)对淋巴细胞增殖的影响。在此基础上,研究不同浓度猪转移因子(transferfactorTF)对犬淋巴细胞增殖的影响。结果表明:在犬淋巴细胞浓度为5×106个/mL,FCS为10%时,PHA-P为12.5μg/mL时,淋巴细胞增殖效果最佳。在最佳培养条件下,猪TF浓度在0.195~25mg/mL时,对犬淋巴细胞转化增殖均有明显促进作用,但猪TF单独或与PHA-P共同作用时对淋巴细胞增殖的最佳刺激浓度均为1.56mg/mL,且猪TF单独作用于淋巴细胞的效应优于与PHA-P的协同作用。试验结果证实,猪TF对犬淋巴细胞增殖有良好的刺激效果。

【Abstract】 the canine bloods with anticoagulant were obtained aseptically and the lymphocytes were isolated. Promoting Effects of different concentration FCS,PBMC and PHA-P on lymphocytes proliferation were tested by orthogonal test. On the above base of the best level combination, different concentration pig TF was investigated to detect its effect on the activation of canine lymphocytes by MTT essay. The result showed that the best levels combination to activate canine lymphocytes proliferation were 2.5×106 canine lymphocyte,6.25 μg/mLPHA-P, 10% calf blood serum, respectively. Under this culture condition, the canine lymphocytes proliferation was remarkable when the pig TF concentration was 0.195 mg/mL~25 mg/mL,but the perfect stimulate concentration of the pig TF separately or combine with PHA-P was 1.56mg/mL. Furthermore, it was confirmed that lymphocytes proliferation under the pig TF separately activation was much better than it cooperated with PHA-P. In the conclusion, the pig TF could promote obviously on canine lymphocytes proliferation and provide a basis for further application of heterogeneous TF on preventing and curing canine’s virus disease.
