聚乙二醇改性壳聚糖固定化L-天门冬酰胺酶的工艺研究

【摘要】 采用交联法制备了聚乙二醇改性壳聚糖(PEG-CS)载体,并将其用于固定化L-天门冬酰胺酶。研究了聚乙二醇浓度、交联剂戊二醛浓度、活化剂甲醛浓度、酶溶液浓度等因素对PEG-CS固定化L-天门冬酰胺酶活力的影响。结果表明:在适当的条件下,PEG-CS固定化L-天门冬酰胺酶的活力可达20~40U/g,并且PEG-CS固定化L-天门冬酰胺酶具有良好的生化性质。

【Abstract】 PEG-CS (chitosan modified by polyethylene glycol), a kind of carrier, was prepared and applied to immobilize L-asparaginase. The preparation and properties of immobilized L-asparaginase on PEG-CS were studied. The effects of reaction process, immobilization time, and concentrations of PEG, cross-linking agent, activation reagent and enzyme solution on the immobilized L-asparaginase were investigated. The results show that, the activity of the immobilized L-asparaginase can reach as high as 20~(40 U/g) under proper conditions for L-asparaginase-immobilization, and the immobilized L-asparaginase possesses good biochemical properties.
