血小板GPⅡb/Ⅲa抗体P140基因的分离及鉴定
Isolation and Identification of Gene Encoding Anti-platelet GPⅡb/Ⅲa Antibody P140
【摘要】 目的:为深入研究抗血小板糖蛋白抗体基因的表达提供途径。方法:采用抑制血小板聚集和间接免疫荧光试验,对3种抗人血小板糖蛋白GPⅡb/Ⅲa单克隆抗体(简称血小板GPⅡb/Ⅲa抗体)P140、P256和H7的抑制血小板聚集的能力和血小板结合特异性进行了比较。将单克隆抗体P140的轻链和重链Fd段基因连接于克隆载体pGEMTEasy上,并进行测序。结果:鼠源血小板GPⅡb/Ⅲa抗体P140的特异性和敏感性优于P256和H7,测序分析结果准确。结论:鼠源血小板GPⅡb/Ⅲa抗体P140血小板结合特异性和抑制血小板聚集的能力均较强。该鼠源性免疫球蛋白基因是一个值得深入探讨的抗血小板抗体基因。
【Abstract】 Objective:To further investigate the expression of the gene of anti-platelet antibody.Methods:Platelet aggregation inhibition test and indirect immunofluorescence assay were employed to compare inhibition against platelet and binding specificity of three mouse anti-human GPⅡb/Ⅲa antobodies,P140,P256 and H7.The genes of Fd fragments and light chain encoding 140Fab antibody were linked to clone-carrier pGEMT-Easy and amplified by RT-PCR from mRNA extracted from hybridoma cell of P140.The genes were sequenced and compared.Results:The specificity of binding to platelet and the ability of platelet aggregation inhibition by mouse anti-P140 antibody were the best among the three antibodies.The sequences of the genes were correct and consistent with Sequences of Proteins of Immunological Interest.Conclusions:The mouse anti-human GPⅡb/Ⅲa monoclonal antibody P140 shown a better specificity of binding to platelet and a better platelet aggregation inhibition action. The mouse antibody gene deserved a further investigation.
