TGF和bFGF对牙周膜细胞ALP活性及Fn合成的影响
Effect of transforming growth factor-α,β and basic fibroblast growth factor on the fibronectin and alkaline phosphatase activity in human periodontal ligament cells in vitro
【摘要】 目的探讨转化生长因子α,β(TGFα,β)和碱性成纤维细胞生长因子(bFGF)对人牙周膜(PDL)细胞碱性磷酸酶(ALP)活性及纤维结合蛋白(Fn)合成的影响。方法体外培养人PDL细胞,与一定浓度的TGFα,β或bFGF作用后,用ELISA法检测Fn含量,用酶动力学方法检测ALP活性。结果TGFα使PDL细胞合成Fn的水平(0.937±0.036)比对照组(0.687±0.023)增加了136.4%(P<0.05),TGFβ使Fn的水平(2.885±0.052)明显提高(P<0.001),bFGF组(0.738±0.041)则没有明显影响(P>0.05)。与对照组(2.431±0.051)相比,TGFβ(1.748±0.042)可抑制PDL细胞的ALP活性(P<0.05),bFGF(1.138±0.046)显著抑制PDL细胞的ALP活性(P<0.01),TGFα(0.331±0.022)的抑制作用更强(P<0.01)。结论TGFα,β和bFGF可能通过调节PDL细胞的ALP活性和合成Fn的水平,发挥PDL细胞在牙周组织的再生中的作用。
【Abstract】 Objective To evaluate biological effects in vitro of transforming growth factor-α,β (TGF-α,β) or basic fibroblast growth factor(bFGF) on the fibronection(Fn) and alkaline phosphatase activity(ALPase) in human periodontal ligament(PDL) cells. Methods PDL cells were obtained from healthy donors. Cells(6×10 5/100 μl) were cultured for 4 days in DMEM supplemented with 1% FBS along(control), or containing TGF-α(10 ng/ml), or TGF-β(3ng/ml), or bFGF(3ng/ml).Fibronectin was determined by ELISA. ALPase activity was measured by enzyme kinetic methods. Data was analyzed by ANOVA. Results TGF-β increased fibronectin levels 4.2-fold. TGF-α caused a slight increase in fibronectin levels(134.6%). bFGF did not influence fibronectin levels. TGF-α and bFGF significantly decreased ALPase activity in PDL cells. TGF-β had less effect than the other peptides. Conclusion The data suggest that these growth factors may be valuable in promoting the regeneration of periodontal connective tissue.
