刺苋花粉特异性变应原成分的分析研究
Analysis of the Specific Antigenic Components of Amaranthus Spinosus
【摘要】 目的:分析刺苋花粉的抗原成分以及刺苋花粉特异性变应原组分,为刺苋花粉变应原的纯化和标准化变应原疫苗的研制提供理论依据。方法:取刺苋花粉浸出液,经十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SD S-PAG E)分离,以16例刺苋花粉过敏的病人血清为探针,进行免疫印迹(W estern b lo tting),检测刺苋花粉浸出液的致敏组分。结果:SD S-PAG E显示可辨蛋白条带有11条,分子量在15~80kD之间,其中主带有9条,分子量依次为18kD、24kD、29kD、35kD、39kD、40kD、42kD、62kD、78kD。W estern b lo tting结果表明,16份刺苋花粉过敏患者的血清全部呈阳性反应,浸出液中共有4条致敏条带,其中主要致敏蛋白的分子量是78kD和40kD。结论:刺苋花粉78kD和40kD的变应原为主要特异性变应原。
【Abstract】 Objective: To investigate the antigenic components of Amaranthus spinosus pollen,and to lay theoretical base for the purification of Amaranthus spinosus pollen allergens and the standardization of allergenic vaccine.Methods: The crude extract was separated by SDS-PAGE.The allergen proteins were analyzed by Western blot with serum obtained from allergic subjects as probes.Results: SDS-PAGE analysis revealed that Amaranthus spinosus pollen proteins comprised at lease eleven discrete protein bands with the molecular weight ranging from 15 kD to 80 kD,nine main bands with the molecular weight being 18 kD,24 kD,29 kD,35 kD,39 kD,40 kD,42 kD,62 kD,78 kD respectively.Western blot assay indicated that the crude extract reacted with serum obtained from sixteen Amaranthus spinosus pollen-allergic subjects and contained four allergen bands in total,and that the 78 kD and 40 kD bands were the major allergenic components.Conclusions: The 78 kD and 40 kD bands were the major allergens of Amaranthus spinosus.
