人卵巢癌细胞系HO-8910裸鼠皮下移植瘤的建立
Subcutaneous Transplantation of Ovary Carcinoma with Cell Line HO-8910
【摘要】 目的:探讨建立卵巢癌细胞系HO8910的裸鼠皮下移植瘤模型的适宜条件。方法:分别用2.5×106/100μL(A组),3.5×106/100μL(B组)和4.5×106/100μL(C组)的细胞接种浓度,注入裸鼠后项中间皮下,观察成瘤情况;用4.5×106/100μL的细胞接种浓度,分别接种于裸鼠后项中间(C组),背部(D组)和前肢腋窝皮下(E组),观察成瘤情况;分别用第38代(F组),第25代(G组)和第10代(C组)的肿瘤细胞4.5×106/100μL,注入裸鼠后项中间,观察各组成瘤情况。结果:A组成瘤率为60%,B组和C组的成瘤率均为100%,但C组的成瘤速度明显高于B组;C组,D组和E组的成瘤率均为100%,注射肿瘤细胞后第2周,C组,D组和E组的肿瘤生长平均相对体积分别为3.0593±0.6851,2.4348±0.4862和2.4039±0.3631,各组间差异无显著性;注射肿瘤细胞后第4周,C组,D组和E组的肿瘤生长平均相对体积分别为13.4832±2.2639,9.3437±1.0986和8.9537±1.0041,C组显著高于D组和E组(P<0.01);G组的成瘤率为30%,F组的成瘤率为50%,与C组相比,差异有显著性(P<0.01)。结论:使用HO8910细胞株,接种浓度为4.5×106/100μL,接种部位为后项中间,接种时尽量使用传代次数少,细胞活性高的细胞,能获得较理想的裸鼠卵巢癌皮下移植瘤模型。
【Abstract】 Objective:To investigate the suitable condition of establishing nude mice model through subcutaneous transplantation with ovary carcinoma cell line HO-8910. Methods: The HO-8910 cells with concentrations of 2.5×106/100 μL(group A), 3.5×106/100 μL(group B) and 4.5×106/100 μL(group C) were injected into nude mice intradermally. The HO-8910 cell concentration of 4.5×106/100 μL was selected, injected into nude mice intradermally(group C),back(group D) and subcutaneous layer of forelimb axillary fossa(group E). The 38th generation(group F), the 25th generation and the 10th generation(group C) tumor cells 4.5×106/100 μL were injected into nude mice intradermally respectively. The growth of tumor was examined in all groups. Results: The rate of tumor formation was 60% in group A and 100% in groups B and C, but the speed of tumor group significantly increased in group C compared with group B; The rate of tumor formation was 100% in groups C, D and E. Two weeks after cell line injection, mean relative tumor volume were 3.0593±0.6851,2.4348±0.4862 and 2.4039±0.3631 in the group C, D and E, with no significantly different among the three groups. Four weeks after cell line injection, tumor mean relative volume of group D and E were significantly less than that of group C [(9.3437±1.0986) and (8.9537±1.0041) versus (13.4832±2.2639)] (P<0.01); The rate of tu formation was 100% in group C, higher than the groups G (30%) and F (50%). Conclusion: Nude mice model of ovary epithelial tumor subcutaneous transplantation was created with HO-8910 cell line; Selection of cells with concentration of 4.5×106100 μL,less passage time and higher viability will help to yield satisfactory rate of tumor formation and optimal animal model of ovary cancer.
