MAGE基因mRNA在非小细胞肺癌中的表达

【摘要】 目的 :检测 MAGE- 1、- 2、- 3、- 4基因 m RNA在非小细胞肺癌 (NSCL C)中的表达情况。方法 :采用逆转录 -聚合酶链反应(RT- PCR)方法对 2 8例 NSCL C患者的肺癌组织进行 MAGE- 1、- 2、- 3、- 4 m RNA表达的检测。结果 :2 8例肺癌标本中 MAGE-1、- 2、- 3、- 4的阳性表达率分别为 35 .7% (10 /2 8)、4 2 .9% (12 /2 8)、5 3.6 % (15 /2 8)、35 .7% (10 /2 8) ;4种基因至少有一种表达的几率是 89.3% (2 5 /2 8) ;两种或两种以上同时表达几率是 5 7.1% (16 /2 8)。非肺癌患者肺组织 4种 MAGE基因表达均为 0 %。结论 :非小细胞肺癌中 MAGE- 1、- 2、- 3、- 4基因高表达 ,非肺癌肺组织无表达 ,提示 MAGE基因可用于肺癌的主动免疫治疗。

【Abstract】 Objective:To investigate the expressions of MAGE-1,-2,-3 and -4 genes in lung cancer samples of patients with non-small cell lung cancer(NSCLC) at mRNA level.Methods:28 samples of lung cancer and 64 stations of lymph nodes from 28 patients with NSCLC were studied to detect mRNA for MAGE-1, -2,-3 and -4 genes by using reverse transcription-polymerase chain reaction assay (RT-PCR).Result:Of the 28 lung cancer samples,the MAGE-1,-2,-3 and -4 were expressed at mRNA level in 35.7%(10/28),42.9%(12/28),53.6%(15/28) and 35.7%(10/28) respectively. At least one of these genes was expressed in 89.3%(25/28) in of the samples. Two or more of these genes were expressed in 57.1%(16/28)of the samples. In the samples of lung of the patients without lung cancer, the MAGE-1,-2,-3 and -4 were expressed at mRNA level in 0%.Conclusion:MAGE-1,-2,-3,-4 are expressed at a high precentage in lung cancer,and all of these genes are negative in the adjacent non-tumorous lung tissue. These results indicate the possiblity of a future specific immunotherapy for lung cancer based on the MAGE antigens.
