水牛β-酪蛋白5′启动区的克隆和序列分析

【摘要】 水牛β-酪蛋白是水牛奶中最重要的蛋白质之一。本实验参照奶牛、绵羊、山羊的β-酪蛋白结构基因序列设计了一对引物P1、P2,以本地水牛基因组为模板,采用高保真ExTaq酶,LA-PCR扩增得到水牛β-酪蛋白基因的5′启动区序列,克隆到pMD18-T上,测序后得到长4.4kb的序列。与牛、山羊和绵羊同区段基因序列进行多重序列比较,并根据多重序列比较结果预测了该区中包含的核心启动子序列TATA盒、5′-UTR(非翻译区)、乳腺组织特异性转录因子(MGF)等重要元件。以该区序列构建的部分反刍动物的进化树与实际的进化关系保持了很好的一致。

【Abstract】 <Abstrcat>Beta-casein is one of the main proteins in the latex of buffalo. A pair of primers was selected to amplify the 5′-regulation segment of buffalo beta-casein gene,which referred the beta-casein gene sequence of bovine,ovine and goat.A 4.4 kb segment DNA was amplified through LA-PCR,with Ex Taq and the genome of native buffalo as template,which was cloned to the pMD-18T and was sequenced thoroughly. The putative binding sites of mammary gland specific transcriptional factors and general nuclear transcriptional factors in 5′-regulation segment of buffalo beta-casein gene were found out by consensus sequence comparison and analysis with other milk genes from bovine and other animal species.The other structure characteristics were also be analyzed.
