固相萃取-反相高效液相色谱法同时测定血清中3种抗癫痫药物

【摘要】 目的 应用固相萃取-反相高效液相色谱法测定血清中抗癫痫药物。方法 用 ElutBond C18小柱预处理样品,甲醇为洗脱剂;采用 Waters Nova RC18 柱,流动相为甲醇∶水=45∶55;检测波长230nm,流速1ml/min,内标(苯乙酮)法定量。结果 苯巴比妥、苯妥英、卡马西平的浓度线性范围分别为4.1~62.6μg/ml(r=0.9968)、3.9~60.1μg/ml(r=0.9971)和 1.6~21.2μg/ml(r=0.9945),最低检测限分别为 0. 05μg/ml、0. 05μg/ml 和 0. 01μg/ml;相对回收率分别为 90. 6% ~103.6%、93.4%~118.3%和91.2%~101.1%;批内变异系数均小于 6.0%,批间变异系数均小于8.0%。结论 固相萃取-反相高效液相色谱法简便、灵敏、准确、稳定,适合于以上 3 种抗癫痫治疗药物的同时监测。

【Abstract】 Objective To establish an advanced method for simultaneous determination of three antiepileptic drugs in serum by solid phase extraction (SPE)-reversed phase high performance liquid chromatography (RP-HPLC).Methods The drugs in serum were enriched by the Elut Bond C 18 (SPE)column,extracted by methanol. The separation was performed on waters Nova R 18 column with mobile phase of methanol:water(45:55,V/V) at a flow rate of 1ml/min and monitored at 230nm. Acetophenone was used as internal standard.Results For aphenyletten,phenytoin and carbamazepine, the linear range was 4.1-62.6μg/ml (r=0.9968),3.9-60.1μg/ml(r=0.9971),1.6- 21.2 μg/ml(r=0.9945) respectively,the detection limit was 0.05μg/ml,0.05μg/ml,0.01μg/ml respectively;the relative recovery rate was 90.6%-103.6%, 93.4%-118.3%,91.2%-101.1% respectively;trecision was within 8%.Conclusion The method presented in this paper is simple, sensitive,stable and suitable to be applied in simultaneous determination of antiepileptic drugs in serum.
