枸杞抗黑果病变异体的初步研究
Elementary Researches on the Resistant Variants of Colletotrichum gloeosporioides Pens
【摘要】 用枸杞品种宁杞1号的髓组织进行离体培养,诱导产生胚性愈伤组织,用110GY(半致死剂量)的60Co-γ射线处理并恢复增殖14d后,以枸杞胶孢炭疽菌(ColletotrichumgloeosporioidesPens)纯菌株产生的培养滤液为选择剂,筛选抗性变异体。在含毒素液60%条件下,一步筛选获得抗炭疽病毒素的愈伤组织,将其转移到含50%毒素液的诱导培养基上培养,经不断选择,选出了抗50%毒素液、且抗性稳定的抗性愈伤组织变异体,该变异体在含30%毒素液的分化培养基中可分化成再生植株,其叶片经胶孢炭疽菌分生孢子接种鉴定,有6株抗病。说明经筛选得到的抗性再生植株是抗炭疽病(黑果病)的变异体。
【Abstract】 At first,embryonic callus from pith culture of Lycium barbarum L.cv.Ningqi No.1 were irradiated with(60 Co-γ) ray at 110 GY(half lethal dose),after 14 days grew normally,the resistant variants will be screened.Resistant callus were screened of cultural filtrate containing 60% toxin of Colletotrichum gloeosporioides Pens.After that,it was medium for inducing callus containing 50% toxin.Plantlet were then regenerated on differentiation medium containing 30% of toxin.Inoculation with suspension of conidium demonstrated that 6 regenerated plantlets were resistant to Colletotrichum gloeosporioides Pens.
