枸杞体细胞胚胎发生及植株再生的研究

【摘要】 以甘肃农业大学林学院实验基地内“宁杞 2 号”枸杞为材料,幼果用 0.1 % HgCl2进行消毒,在无菌条件下取出未成熟胚进行诱导,在胚成苗后进行继代和生根培养。培养结果表明:以 MS 为基本培养基,附加 6-BA1.0 mg/L和 IBA0.5 mg/L对体细胞胚胎诱导最有效;在 2,4-D为 1.0 mg/L和 1.5 mg/L时的培养基上胚性愈伤组织诱导率较高,分别为 51.0 %和 56.7 %;黑暗低温(10℃)处理 5d 不但可以促进细胞胚胎的发生,而且可以提高其发生的频率(60.0%)。

【Abstract】 Cultivars Ningqi No.2 was used as the materials. An immature pterodia was taken from young fruit disinfected with 0.1% HgCI2. Shoot multiplication culture and rooting culture were conducted after emergence of seedlings. The result indicates that the induction of somatic embryos showed the best when the embryonic callus were cultured on MS medium supplemented with 1.0 mg/L 6-BA and 0.5 mg/L IBA. The embryogenic callus (EC) frequency was 51.0 % and 56.7 %at 1.0 and 1.5 mg/Lof 2,4-D added in MS medium, respectively. Dark and low temperature (10 ℃) for 5 days promoted development of somatic embryogenesis and increased its frequency (60.0 %).
