肾细胞癌中P-gp、GST-л和TOPOⅡ的表达及其意义
Expression and significance of P-gp, GST-л, TOPOⅡ gene protein in renal cell carcinoma
【摘要】 目的:探讨多药耐药(MDR)基因产物P糖蛋白(P- gp)、谷胱甘肽- S-转移酶(GST- л)和DNA拓扑异构酶 (TOPO )在肾细胞癌(renal cell carcinoma,RCC)组织中的表达及临床意义。方法:采用免疫组化S- P方法检测4 5例肾细胞癌组织和1 0例正常肾组织中P- gp、GST- л和TOPO 蛋白的表达情况。结果:1 90 %的正常肾组织近曲小管上皮细胞膜和/胞质有P- gp表达;P- gp、GST- л和TOPO 蛋白在肾细胞癌组织中的阳性表达率分别为77.8%,71 .1 %和31 .1 %。P- gp、GST-л和TOPO 阳性表达与肾细胞癌组织学分型、组织学分级、肿瘤大小及有无脉管浸润转移均无关(P>0 .0 5 ) ;2两种MDR基因产物的共表达率为5 1 .1 %,两种或两种以上MDR基因产物的共表达率为71 .1 %,均明显高于单独基因的表达率1 5 .6 %(P- gp)和8.9%(TOPO )。GST-л表达均伴有P- gp和/或TOPO 的表达;P- gp表达和GST- л表达呈明显正相关(P<0 .0 1 )。TOPO 表达与P- gp、GST- л表达均无相关性(P>0 .0 5 )。结论:P- gp、GST- л蛋白高表达和TOPO 蛋白低表达在肾细胞癌的多药耐药中起重要作用,多基因联合检测有助于判断术后化疗疗效。
【Abstract】 Objective:To explore the expression and clinical significance of P-gp, GST-л and TOPOⅡ protein in human renal cell carcinoma. Methods:The expression of P-gp, GST-л and TOPO Ⅱ protein were detected in 45 cases of renal cell carcinoma tissue with S-P immunohistochemical method. Results:①the positive expression of P-gp was 90% in kidney tissue, and that of P-gp, GST-л and TOPOⅡ separately were 77.8% 71.1% and 31.1% in RCC. No significant relationship was found between the positive expression rates of GST-л, P-gp and TOPO Ⅱ protein and the size, histopathological classification in RCC(P>0. 05).②The coexpression rate of only 2 MDR gene production and only 2 or 3 MDR gene production separately were 51.1% and 71.1%, which exceeded apparently more than individual expression rate 15.6%(P-gp) and 8.9%(TOPOⅡ); P-gp significantly positively correlated with GST-л; P-gp and GST-л separately had not obviously negative correlation with TOPOⅡ. Conclusion:The high-expression of P-gp, GST-л and the lower-expression of TOPOⅡ in RCC play an important role in influence of tumor MDR.
