胰酶制备新工艺的研究
Preparation of pancreatin from swine pancreas
【摘要】 目的研究从猪胰腺中提取胰酶的新工艺。方法使用φ=7. 5%异丙醇为提取溶剂,添加ρ=0. 2% CaCl2 和φ=1%胰酶粉为激活剂以及适量的保护剂,并加入NaHCO3 使体系的pH稳定在 6. 5 ~7. 0之间,在室温下提取 3h。结果在优化工艺条件下,制得的胰酶制品中的胰蛋白酶、胰淀粉酶、胰脂肪酶的酶活力分别达 5 200、57 300、37 000u/g以上,为中国药典所规定的最低酶活的 8~10倍,收率 10% ~12%。结论该工艺简单、提取效率高、产品酶活高。
【Abstract】 Objective To study the new processing technique of pancreatin from swine pancreas. Methods Used 7.5% isopropyl alcohol as extracting solvents; added 0.2% CaCl 2 + 1% pancreatin as protecting agents; regulated pH 6.5~7.0 with NaHCO 3; extracting time 3 h. Results Under optimum conditions, pancreatin with high activity and yield was obtained. The activities of trypsin, lipase and amylopsin were over 5 200 u/g, 57 300 u/g and 37 000 u/g respectively, and the yield was 10%~12%. Conclusion It is a simple and time saving technique for the preparation of pancreatin with high purity, high activity and high yield.
