广东省韶关地区戊型肝炎病毒基因型分析
Phylogenetic analysis of hepatitis E virus (HEV) in fecal samples from pigs and in serum from human in Shaoguan ,Guangdong province
【摘要】 目的了解韶关地区流行的戊型肝炎病毒基因型,并探讨戊型肝炎是否属于人兽共患病。方法通过同源比对HEV四种基因型的戊型肝炎病毒序列,选取其保守序列部分设计两对巢式聚合酶链式反应的简并引物,建立一种可同时扩增戊型肝炎四个基因型病毒RNA的PCR快速检测方法。在广东省韶关地区采集猪粪便标本和非甲非乙型肝炎病人血清,提取病毒RNA,然后进行PCR检测,阳性时对扩增片段进行基因序列测定,并进行基因型分析。结果RT-nPCR检测结果显示99份猪粪便标本中有6份HEVRNA阳性,7份病人血清样本中1份阳性,序列分析显示全部为HEV基因IV型,4份猪标本中的HEV基因序列与GenBank中人HEV的基因序列最相似,同源性为88%~94%,且其中1份猪标本中的HEV与1份病人标本中HEV的序列同源性高达91.5%。结论广东韶关地区HEV流行株为基因IV型,而且猪HEV基因序列与人HEV之间有着高度的相似性,结果证实戊型肝炎是一种人兽共患传染病。
【Abstract】 ObjectiveTo investigate the genotyping of HEV in Shaoguan, Guangeong province. MethodsTwo pairs of consensus primers were designed according to the conserved sequence region from four-genotype HEV strains for nested PCR. HEV RNA in pig fecal sample and in serum from patient with non-A, non-B hepatitis was detected by nested PCR. Positive PCR product was sequenced and analyzed phylogenetically with neighbor-joining method. ResultsHEV RNA was detected in 6 of the 99 pig fecal samples and 1 of the 7 patient sera. Sequence analysis show that all of the positive isolates were genotype IV. ConclusionThese findings suggest that HEV genotype IV is the endemic strain in Shaoguan and hepatitis E is a zoonosis.
