【摘要】 目的检测白细胞介素-2(IL-2)联合IL-12诱导人外周血单个核细胞(PBMC)体外抗肿瘤作用。方法分别应用MTT法和流式细胞仪法测定IL-2与IL-12协同诱导对体外培养的3种肿瘤细胞株的杀伤活性。结果IL-2与IL-12联合诱导的PBMC,对3株靶细胞的杀伤效应差异有显著性(P<0.05)。MTT法与流式细胞仪法两种方法检测的杀伤率有明显的相关性(r=0.997,P<0.05)。结论IL-2与IL-12协同对3种不同组织来源的实体瘤细胞株表现了较强的杀伤力,而杀伤率的差异与肿瘤细胞的不同免疫特征有关。更多还原

【Abstract】 Objective To examine the in vitro antitumor activity of normal human peripheral blood mono-nuclear cells(PBMC) induced by interleukin-2(IL-2) and IL-12.Methods MTT and flow cytometry assay were used to detect the cytotoxic effects of IL-2 or/and IL-12 on 3 cell lines.Results PBMC induced by IL-2 and IL-12,had significantly different cytotoxic effects on three cell lines(P<0.05).A good correlation was observed between the cytotoxicity by MTT and flow cytometry(r=0.997,P <0.05).Conclusion The cytotoxic synergy by IL-2 and IL-12 against 3 tumor cell lines in vitro is significantly differents.The different antitumor effects maybe associated with different immune characteristics of tumor cells.更多还原
