1-羟基-2-(5-NO2-2-吡啶偶氮)-8-氨基-3,6-萘二磺酸光度法测定血清白蛋白的研究

【摘要】 研究了新显色剂1 羟基 2 (5 NO2 2 吡啶偶氮) 8 氨基 3,6 萘二磺酸(简称5 NO2 PAH)与牛血清白蛋白(BSA)的显色反应,建立了5 NO2 PAH光度法测定血清蛋白的新方法。结果表明,在pH3.0的B R缓冲液中,牛血清白蛋白与5 NO2 PAH定量络合,导致5 NO2 PAH在530nm处的吸收降低,且吸光度降低值与BSA量在6~31μg mL范围内呈线性关系,摩尔吸光系数为2 2×105L·mol-1·cm-1,方法检出限为9.8×10-8mol L。用摩尔比法测得结合数为130。
【Abstract】 A new method for determination of bovine serum albumin (BSA) by spectrophotometry based on the colour reaction between BSA and the new chromogenic reagent 1-hydroxy-2-(5-nitro-2-pyridylazo)-8-aminona phthalene-3,6-disalfonic acid (5-NO2-PAH) has been developed. The experimental results show that in a Britton-Robinson buffer solution of pH 3.0 and at room temperature, BSA can react with 5-NO2-PAH to form a stable complex and cause a decrease of absorbance for 5-NO2-PAH itself at 532 nm. The decrease of absorbance was linear with the concentration of BSA over the range of 6~31 μg/mL. The detection limit of BSA was 9.8×10-8 mol/L. The method has also been applied to the determination of BSA in human serum with satisfactory results.
