极低密度脂蛋白对大鼠系膜细胞分泌FN以及TGFβ1mRNA转录的作用
Effect of very low density lipoprotein on secretion of fibronectin and gene transcription of transforming growth factor-beta 1 of rat mesangial cells
【摘要】 目的观察极低密度脂蛋白对大鼠系膜细胞分泌FN以及对转化生长因子- β1 (TGF- β1) 基因转录的作用, 进而探讨 VLDL对系膜细胞的毒性作用。方法应用 ELISA法测定培养细胞上清纤维连接蛋白 (Fi bronectin, FN) 的分泌; RT- PCR法测定系膜细胞 TGF- β1 的基因表达情况。结果在一定的时间范围内 (6 ~24 h),VLDL可以以时间、剂量依赖性的关系促进系膜细胞分泌FN。随着刺激时间的增加, TGF -β1mRNA转录逐渐增强; VLDL 浓度在 0 ~ 500μg/ml 之间时, TGF- β1mRNA 转录随着 VLDL 浓度的增加而逐渐增强。结论VLDL可以促进系膜细胞分泌FN以及TGF- β1 的基因转录, 提示VLDL具有肾脏毒性。
【Abstract】 Objective To observe the effect of very low density lipoprotein on secretion of fibronectin and transforming growth factor-beta 1 gene transcription of rat mesangial cells and to study if VLDL has nephrotoxicity on mesangial cells.Methods ELISA method were employed to determine protein production of fibronectin.RT-PCR method was adopted to determine mRNA transcription of TGF-beta 1.Results Within range of concentration(0~500 μg/ml),VLDL could promote fibronectin production with time and dose dependent manners.Within rather wide range of concentration(10~500 μg/ml),mRNA transcription of TGF-beta 1 were stimulated with time and dose dependent manners.Conclusion VLDL would promote fibronectin production and mRNA transcription of TGF-beta 1 of rat mesangial cells.VLDL may has nephrotoxity.
