【摘要】 目的探讨黏附分子CD44s在体外培养人眼小梁细胞(HTC)的表达及其与小梁细胞结构和功能的关系。方法对体外培养的HTC应用流式细胞术和RTPCR法检测CD44s的表达。结果HTC高表达CD44s黏附分子,阳性细胞数达92%以上,而且均表达CD44smRNA。结论体外培养的HTC表达CD44s黏附分子。CD44s与其配体透明质酸(HA)的协同性调节HTC的内环境并在维持其结构和功能方面起重要作用。

【Abstract】 Objective To study the expression of adhesion molecule CD_ 44s in cultured human trabecular meshwork cells(HTC) and to discuss their possible relationship between structure and function of HTC. Methords HTC were cultured in DMEM/F12 medium. CD_ 44s was labelled with fluorescein isothiocyanate(CD_ 44s-FITC) and incubated with the HTC . Total RNA from the cells were extracted with Trizol reagent. CD_ 44s mRNA exprssion in the HTC was examined by using reverse transcription polymerase chain reaction(RT-PCR). Results Positive expression of CD_ 44s was observed in the cultured cells with flow cytometry. Positive cell number reached over 92%,all expressed CD_ 44s mRNA. Conclusion CD_ 44s is present in the cultured HTC. Cooperativity between CD_ 44s and HA modulate the internal environment of trabecular meshwork cells and maintains structure and function.
