碱性成纤维细胞生长因子诱导大鼠缺血心肌血管新生的作用
Basic Fibroblast Growth Factor Induces Myocardial Angiogenesis in Rat Model of Ischemia
【摘要】 目的 探讨局部应用碱性成纤维细胞生长因子(bFGF)对大鼠急、慢性缺血心肌血管新生的作用。 方法 健康雄性Wistar大鼠,采用开胸结扎左冠状动脉前降支法制备急、慢性心肌梗死模型,并随机分为bFGF组和对照组(n =8) ,bFGF组以0 .0 2mL(2 0 μg/kg ,0 .2 5mg/mL)造模时或造模1个月后于结扎处局部注射,对照组以等容积生现盐水处理。注射后1个月观察血流动力学参数、心肌梗死面积和侧支微血管数目的变化。 结果 bFGF组心率、左室舒张末压明显低于对照组(P <0 .0 1)、平均动脉压、左室收缩压、左室最大收缩速率明显高于对照组(P <0 .0 1) ;bFGF组心肌微血管数明显多于对照组(P <0 .0 1) ;急性心肌梗死模型bFGF组心肌梗死面积明显小于对照组(P <0 .0 1) ,慢性心肌梗死模型bFGF组心肌梗死面积与对照组比较差异无显著性(P >0 .0 5 )。 结论 局部应用bFGF能明显促进缺血心肌血管新生,缩小急性心肌梗死面积,改善血流动力学。
【Abstract】 Objective To study whether basic fibroblast growth factor(bFGF) can induce myocardial angiogenesis in a rat model of acute or chronic myocardial ischemia. Methods 36 male Wistar rats underwent thoracotomy and ligation of left anterior descending coronary artery. After acute and chronic myocardial infarction model reproducing, bFGF group or saline solution(control group) were injected into border myocardium between infracted and mon-infarcted areas. One month later, hemodynamic parameters, microvessel number in the infarct size were observed. Results Heart rate(HR) and left ventricular end diastolic press(LVEDP) were significantly lower in bFGF group than that in control group(P<0.01), MAP, LVSP, +dp/dt max, -dp /dt max of bFGF group were significantly higher than that of control group(P<0.01). Capillary density was significantly higher. The infarct size of bFGF group was significantly smaller than that of control group in a rat model of acute myocardial ischemia. Although chronic myocardial ischemia no any significant difference in the infarct size was found. Conclusion Local usage of bFGF can promote ischemic myocardial neovascularization, therefore reduce acute infarct size and improve hemodynamics of infarcted hearts.
