透明质酸对小鼠卵丘细胞外基质的影响

【摘要】 目的 观察透明质酸 (HA )对小鼠卵丘细胞外基质的影响。 方法 取 3~ 4周龄的雌鼠 (含野生型和bikunin基因敲除小鼠 )诱导排卵 ,取卵丘 -卵母细胞复合体 (COC) ,镜下计数诱导排卵后卵母细胞。免疫荧光双重染色后观察 HA及血清来源的 HA相关蛋白 (SHAP)在卵丘细胞外基质的分布。 结果  SHAP- HA复合体缺失引起卵丘细胞外基质的变化 ,并影响到随后的排卵。 结论  SHAP- HA参与卵丘细胞外基质的构建 ,细胞外基质的受损引起卵母细胞功能的破坏。

【Abstract】 Objective To study the effects of hyaluronan(HA) on the cumulus extracellular matrix in mice. Methods Three-to four-week-old female mice(wild-type and bikunin-null mice) were instimulated to superovulation and isolation of cumulus oocyte complex(COC). The COC were examined with microscope. The distribution of HA and serum-derived hyaluronan-associated proteins(SHAP) in the cumulus extracellular matrix detected with immunofluoresence staining method. Results SHAP-HA complex-deficiency may causes abnormality of the cumulus matrix,and subsequently affect the successful ovulation. Conclusion SHAP-HA complex is required for the construction of intercellular matrices; the function of the oocytes were reduced significantly for the malformation of the matrices.
