利用tyrB与aspA基因在大肠杆菌中的偶联表达制备L-苯丙氨酸(英文)

【摘要】 报道了利用tyrB与aspA串联表达来合成苯丙氨酸.首先将tyrB与aspA串联在质粒pBV221上,构成串联表达质粒pBV-tyrB-aspA;然后将该重组质粒转化到E.coliK36菌株中表达.对基因编码的相关酶活性以及工程菌利用FA和PPA生成Phe进行研究,结果表明:利用AspA和TyrB的偶联反应可以有效的提高E.coliK36合成L-苯丙氨酸的能力.

【Abstract】 To improve productivity,a special biosynthetic route has been applied in the phenylalanine-producing strain Escherichia coli K36.By coupling tyrB and aspA,the biosynthesis of phenylalanine was completed in the reaction solution containing compounds PPA,FA and NH+4.Here,gene tyrB and aspA of E.coli K12 were inserted into plasmid pBV221 to construct a recombinant plasmid pBV-tyrB-aspA,and it was introduced into E.coli K36.The enzymatic activities of AspA and TyrB were measured and the phenylalanine productivities of the recombinant strains were analyzed.The results demonstrated that co-expression of tyrB and(aspA) was able to enhance phenylalanine biosynthesis.
