SD大鼠超数排卵及早期胚胎体内发育

【摘要】 60只雌鼠随机分为3组,第1组为PMSG+hCG处理组,第2组为LHRH-A2处理组,第3组为自然发情组;于处理大鼠见栓后第0.5天至第5天从输卵管或子宫中回收胚胎。结果表明,第2组每只大鼠平均回收胚胎10.07枚±3.81枚,优于第1组(平均回收8.71枚±1.13枚胚胎),但两组间无统计学差异(P>0.05);两个超排处理组的回收胚胎数均明显高于自然发情配种组(平均回收胚胎6.27枚±3.23枚,P>0.05)。自然发情大鼠在见栓后0.5 d~1 d,1 d~1.5 d,1.5 d~2 d,2 d~2.5 d,2.5 d~3 d,3 d~3.5 d和3.5 d~4.5 d,胚胎分别发育到1-细胞期,1-细胞至2-细胞期,2-细胞期,2-细胞至4-细胞期,4-细胞至8-细胞期,8-细胞期,桑椹胚和囊胚期;自然发情与超排处理的SD大鼠,其早期胚胎发育阶段与进入子宫的时间不尽相同。自然发情大鼠的胚胎多在近桑椹胚期进入子宫,而超排大鼠在8-细胞期即已进入子宫。该结果为进一步开展相关研究奠定了基础。

【Abstract】 A total of 60 female rats were randomly divided into three groups.Group I was treated with PMSG+ hCG,Group Ⅱ was injected with LHRH-A2and Group Ⅲ was natural estrous and bred control.Embryo collection was conducted via flushing the oviducts or uterus of donor rats on the day 0.5-5 after the appearance of vaginal plug.Group Ⅱ and Group Ⅰ in the average number of 10.07±3.81 and(8.71±)1.13 embryos respectively,which were significantly higher than the average number of 6.62±3.23 embryos from the control(P>0.05).Our results showed that the 1-cell stage,1-to 2-cell stage,2-cell stage,2-to 4-cell stage,4-to 8-and 8-cell stage,morula and blastocyst could be reached on the day 0.5-1,1-1.5,1.5-2,2-2.5,2.5-3,3-3.5,and 3.5-4.5,respectively,for natural estrous rats.There were exist some differences in the developmental stages of embryos and the time-point of entering uterus between natural estrous and superovulated rats.Most embryos from natural estrous rats migrated into the uterus ahead of morula stage,while embryos from superovulated rat entered the uterus at 8-cell stage.These results established the basis for further related studies.
