【摘要】 根据测定的猪繁殖与呼吸综合征病毒广东株(PRRSV GD)的序列,设计一对引物。用保存的PRRSV GD 的ORF5 的克隆产物pMD ORF5 质粒作为模板,扩增ORF5基因。将该片段定向插入到原核表达载体pBAD/gⅢC中,转化大肠埃希菌。重组质粒经PCR和酶切鉴定后,用阿拉伯糖诱导表达。用Western blotting鉴定表达蛋白,证明ORF5基因得到表达。更多还原

【Abstract】 The ORF5 gene of Porcine reproductive and respiratory syndrome virus was amplified by polymerase chain reaction (PCR).The DNA fragment was ligated with plasmid pBAD/gⅢC and transformed into E.coli.The constructed recombinant plasmid was confirmed by RE digestion and sequencing, and induced with arabrose.
