微卫星PAGE银染法及其干胶的简易制备
Sliver Staining of Microsatellite DNA-PAGE and Simpe Method of Making Dry Gel
【摘要】 为了使微卫星PCR扩增的DNA片段能得到较好的分离和长期保存实验胶片 ,研究利用 1 5对猪的微卫星引物 ,经PCR扩增 ,非变性聚丙烯酰胺凝胶电泳 ,银染法显色 ,然后制成了干胶。结果表明 ,聚丙烯酰胺凝胶具有很好的分离微小差异DNA片段的能力 ,而且银染后制成的干胶可长期保存。此法尤其适用于不具备凝胶成像系统和干胶机的实验室使用。
【Abstract】 In order to fully separate DNA fragments after polymerase chain reaction(PCR)and enduringly preserve PAGE gel, 15 microsatellite primers on swine were used during expriment. First, making PCR; then, the fragments were separated by polyacrylamide gel electrophoresis (PAGE)and appeared by silver staining; last, the gels were processed for dry gels.The result indicated PAGE had very good effect on separating the discrepancy DNA fragments only few base pairs.The dry gel can be preserved for ever.This method especially apply for some laboratories without gel imaging system and gel drier.
