重组PP150抗原用于胶体金免疫斑点技术检测抗HCMV-IgM
Recombinant PP150 of human cytomegalovirus as antigene for colloidal gold immuno-dot assay to detect HCMV-IgM in sera
【摘要】 目的:探索应用重组人巨细胞病毒(HCMV)PP150抗原用于免疫滴金技术检测血清中抗HCMV-IgM的诊断价值.方法:PCR扩增HCMV pp150抗原决定簇(840~1048 aa)编码基因片段,插入原核表探索应用重组人巨细胞病毒(HC-MV)PP150抗原用于胶体金免疫斑点技术(CG IDA)检测血清中抗HCMV-IgM的诊断价值.达载体pMA l-p2,转化宿主菌E.coli,诱导表达及纯化.重组抗原CG IDA与酶联免疫吸附试验(ELISA)试剂盒对比检测.结果:重组抗原CG IDA与ELISA试剂盒比较,符合率达96.0%;CG IDA检测HCMV-IgM的敏感度为74.2%,特异性为100%.结论:重组抗原pp150用于CG IDA对HCMV的早期诊断,具有较好应用价值.
【Abstract】 AIM: To use recombinant pp150 of human cytomegalovirus(HCMV) in colloidal gold immuno-dot assay(CGIDA) for the detection of HCMV-IgM in sera.METHODS: A fragment of gene coding for pp150 antigen epitopes(840-1048aa) was expressed in E.coli and the fusion protein was purified.The results were compared with those of specific antibodies ELISA IgM.RESULTS: Recombinant antigen CGIDA had a good agreement(96.0%) with ELISA kit.The specificity of CGIDA was 100% and the sensitivity was 74.2%.CONCLUSION: Recombinant pp150 is an effective and valuable antigen used in CGIDA for detecting HCMV-IgM.
