宁夏回族5个Y-STR基因座及单倍型遗传多态性分析

【摘要】 目的:研究宁夏回族5个YSTR基因座及单倍型遗传多态性,估价它们在法医学中的应用价值和丰富中华民族基因信息资源.方法:调查宁夏回族101个无关、健康男性个体.研究5个YSTR基因座基因及单倍型频率分布.每个基因座的引物分别用FAM,HEX或TAMRA荧光标记,通过复合扩增、基因扫描、基因分型和遗传多态性分析获得数据.结果:5个YSTR基因座共检测到29个等位基因,频率分布在0.0099~0.5446.101个个体共检测到94个单倍型,87个是唯一的,有7个单倍型被检测2次.单倍型的多样性为0.9986.结论:该数据可以很好的应用于法医学个体识别和亲权鉴定以及群体遗传学研究.

【Abstract】 AIM: To study genetic polymorphisms of 5 Y chromosomal STRs in Hui ethnic group living in Ningxia Hui Autonomous Region and to assess their value in forensic science. METHODS: We investigated 101 unrelated, healthy, male individuals of Hui ethnic group and studied their allelic frequency distributions and haplotype diversity of 5 Y chromosomal STRs. Primer for each loci was labeled with the fluorescence, such as FAM (blue), HEX (green) or TAMRA (yellow). The data of Hui ethnic group were obtained through co-amplification, GeneScan, genotyping, and genetic distribution analysis. RESULTS: Twenty-nine alleles were detected, with frequencies ranging from 0.0099 -0.5446. Out of the total of 101 individuals, 94 were of different haplotypes, 87 were unique and 7 were found 2 times. The haplotype diversity for 5 Y-STR loci was 0.9986. CONCLUSIONS: The data can be well useful for individual identification and paternity testing in forensic fields and in the study of population genetics.
