小鼠帕金森病模型中的大脑皮质细胞因子表达
Cytokine alteration of cerebral cortices in murine model of Parkinson’s disease
【摘要】 目的:探讨甲基苯基四氢吡啶(MPTP)是否可诱导小鼠大脑皮质内细胞因子水平变化.方法:28只C57BL/6J雄性小鼠接受MPTP25mg/kgip注射连续5d,对照组(10只)注射生理盐水,在最后一次注射的第1日(10只),第3日(8只)和第14日(10只)杀鼠,然后用酶联免疫吸附的方法检测大脑皮质匀浆液上清内细胞因子IL1β和IL6的水平.结果:注射MPTP对大脑皮质IL1β水平有影响(P<0.001),与对照组相比,IL1β水平在注射后第3日降低(P<0.01);MPTP注射对大脑皮质IL6水平没有影响.结论:MPTP诱导了大脑皮质促炎症细胞因子IL1β的水平改变.
【Abstract】 AIM: To explore the effects of 1-methyl-4-henyl-tetrahydropyridine (MPTP) on cerebral cortical cytokines. METHODS: Twenty-eight male C57BL/6J mice were treated with 25 mg/kg MPTP i.p. for five consecutive days. The control group (10 mice) were treated with saline. The mice were decapitated on day 1 (10 mice), day 3 (8 mice) and day 14 (10 mice) after the last MPTP injection. IL-1β and IL-6 levels in supernatant of homogenates of cerebral cortices were measured by the enzyme linked immunosorbent assay (ELISA). RESULTS: MPTP injection affected cerebral cortical IL-1β levels (P<0.001) and compared with those in control group, IL-1β levels decreased dramatically at day 3 after treatment (P<0.01). MPTP injection had no effect on cerebral cortical IL-6 levels. CONCLUSION: MPTP induces IL-1β variations in cerebral cortices.
