纳洛酮对离体心肌缺血再灌注损伤大鼠心脏功能的影响
The Effects of Naloxone on Cardiac Function during Myocardial Ischemia-reperfusion Period
【摘要】 目的:在离体大鼠心肌缺血再灌注模型上,观察阿片受体特异拮抗剂纳洛酮对大鼠心功能的影响,并探讨其机制。方法:建立改良的Langendorff离体大鼠心脏灌注模型,缺血40min,再恢复常速灌注30min,造成心肌缺血再灌注损伤。用两种不同浓度的纳洛酮(16mg/kg和32mg/kg)分别灌注缺血的心脏后,观察再灌注15min时①心脏左室内压(LVP)、左心室内压最大变化率(±LVdly/dt)、心率(HR)的变化;②冠脉流量、冠脉流出液中总蛋白、肌红蛋白含量、乳酸脱氢酶(LDH)的变化。结果:用纳洛酮灌流缺血的心脏后,与单纯缺血再灌注组比较:①左室内压、左心室内压最大变化率(±LVdp/dt)分别增加;②平均冠脉流量增加、冠脉流出液中总蛋白、肌红蛋白含量和乳酸脱氢酶(LDH)含量均明显减少。结论:纳洛酮对离体的缺血再灌注损伤心脏有保护作用。该研究拓宽了纳洛酮的临床用途,为防治缺血性心脏病提供了理论依据。
【Abstract】 Objective: To investigate if Naloxone is concerned with cardiac ischemia-reperfusion ( I/R) process and to study its effects on cardiac function Method : A Langendorff rats model were built, then isolated rat hearts were subjected to ischemia for 40 min and reperfusion for 30 min .Results: when Naloxone ( 16mg/kg and 32mg/kg ) was given in reperfusion period. (1)In isolated perfused rat heart models,±dp/dt max of the heart increased; (2) The average volume of coronary efflux increased respectively, compared with that of control group of ischemia-reperfusion , and the amount of total protein, myoglobin and activities of LDH in effluent samples of Naloxone group were respectively lower than group of ischemia-reperfusion. Conclusion To some extent, Naloxone may have myocardial protective effects during ischemia-reperfusion period.
