复合酶法提取姬松茸胞内多糖
Extraction of Intracellular polysaccharides from Agaricus blazei Murill by compound hydrolyzation
【摘要】 以液体深层发酵培养的姬松茸菌丝体为原料,利用酶法提取姬松茸胞内多糖。通过单因素试验、正交试验及方差分析确定了复合酶法提取胞内多糖的最佳条件:pH 4.5、温度45℃、时间1.5 h、木瓜蛋白酶1.0%、纤维素酶0.5%、果胶酶1.0%。在此条件下,胞内多糖的提取率为3.70%。胞内多糖经Sephadex G-200柱层析后得到两种多糖组分,经测定此两种多糖组分的相对分子质量分别为91 000和40 500。
【Abstract】 The intracellular polysaccharides of Agaricus blazei Murill by submerged culture were extracted by enzyme hydrolysis method.By single factor tests,orthogonal experiments,and variance analyses,the optimum hydrolytic condition is determined as follows: pH value 4.5,temperature(45℃,) time 1.5h,papain 1.0%,cellulase 0.5%,and pectinex 1.0%.Under this condition,the yield of intracellular polysaccharides is up to 3.70%.Two kinds of polysaccharides are obtained by Sephadex G-200 column chromatography with molecular weight 91000 and 40500,respectively.
