酶解蚕蛹蛋白工艺的研究及产物分析
Study on technology for hydrolysis of chrysalis’s protein and its products’ analysis
【摘要】 报道了酶法水解蚕蛹蛋白工艺的实验研究,并对水解产物进行了分析。实验中选用已确定的中性蛋白酶和动物蛋白酶对蚕蛹蛋白水解。结果表明,最佳酶解条件为:温度50℃,pH7.0,底物蚕蛹蛋白的质量分数1.0%,动物蛋白酶加量150U/g,中性蛋白酶与动物蛋白酶酶活比为4∶1,水解时间3h,在此条件下水解度为27.36%,酸溶性肽得率为55.17%,相对分子质量分布于336~638。
【Abstract】 This paper reports the technology investigation for hydrolysis of chrysalis’s protein and the analysis of its products. In the experiment, the hydrolysis of chrysalis’s protein is performed by using the method of double enzyme hydrolysis with determined Neutrase and animal protease. The suitable enzyme hydrolysis condition is determined as follows: teparature, 50?℃;pH, 7.0, substrate concentration, 1.0%; ratio of animal protease enzyme/substrate, 150?U/g; Neutrase/animal protease, 4; hydrolysis period of time, 3.0?h. The degree of hydrolysis is 27.36%. The yield of acid soluble peptide is 55.17%. The molecular weight of poly-peptide is about 336 to 638.
