小剂量60Co放疗对血管纤粘连蛋白及细胞基质金属蛋白酶-2基因表达的影响
The Effect of 60Co Brachytherapy on the Gene Expession of Fibronectin and MMP-2
【摘要】 目的 探讨小剂量60 Co放射治疗对血管纤粘连蛋白和细胞基质金属蛋白酶 -2基因表达的影响及其机制。方法 以0、7、14、2 8Gy60 Co射线辐射培养的大鼠主动脉平滑肌细胞 ,观察其对纤粘连蛋白 (fibonectin ,FN)及细胞基质金属蛋白酶 -2 (ma trixmetalloproteinase -2 ,MMP -2 )mRNA表达的影响。结果 7Gy放疗组对细胞MMP -2及FNmRNA表达与对照组比较无显著差异 ,而 14Gy、2 8Gy放疗组FNmRNA分别为 72 0 5± 5 82和 49 77± 5 62 ,较对照组 (95 17± 6 41)比较有显著差异 (P <0 0 5 )。MMP -2mRNA在 14Gy、2 8Gy放疗组分别为 64 2 5± 3 81和 42 17± 3 16,较对照组 (82 5 2± 6 5 0 )比较有显著差异 (P <0 0 5 )。结论 14、2 8Gy60 Co射线辐射可以抑制MMP -2合成和FN等胞外基质的代谢。
【Abstract】 Objective To study the effect of 60 Co radiotherapy on the gene expession of fibronectin (FN) and matrix metalloproteinase-2 (MMP-2) in the cultured rat vascular smooth muscle cells (VSMCs). Methods Rat VSMCs were cultured in DMEM containing 10% FBS, and were radiated by 60 Co at the doses of 0,7,14 and 28Gy, respectively. mRNA level of FN and MMP-2 was measured by RT-PCR. Results The expression level of MMP-2 mRNA decreased from 82.52±6.50 to 64.25±3.81 and 42.17±3.16 in VSMCs with 14,25Gy radiation, respectively(P<0.05). The expression level of FN mRNA decreased from 95.17±6.41 to 72.05±5.82 and 49.77±5.62 in VSMCs with 14,25Gy radiation, respectively(P<0.05). Conclusion 60 Co radiotherapy inhibited the expression of MMP-2 and FN in VSMC cells.
