磺胺二甲基嘧啶人工抗原合成及多克隆抗体制备
Preparation of artificial antigen and polyclonal antibody for sulfamethazine immunoassay
【摘要】 磺胺二甲基嘧啶(SM2)为只具有反应原性而无免疫原性的半抗原。本试验将SM2重氮化后分别连接人血清白蛋白(HSA)、卵清蛋白(OVA),合成人工抗原,经紫外光谱扫描验证偶联结果,计算结合比分别为4∶1、3∶1。用偶联物SM2-HSA免疫新西兰白兔,以SM2-OVA为包被原,4次免疫后ELISA方法测定抗血清效价为1∶12 800。表明抗原合成成功,这为小分子药物残留的免疫学检测奠定了技术基础。
【Abstract】 Sulfamethazine(SM2) is a small molecular hapten possessing immunoreactivity but not possessing immunogenicity.It was coupled with human serum albumin(HSA) and ovalbumin(OVA) by diazotization.The complete antigens were identified by UV scan,and the molecular ratio of SM2 to HSA and OVA were 4 and 3.The conjugate of SM2-HSA was used to immunize rabbits.After 4 times of booster injection,the produced antiserum showed special sensitivity and high titer to the Sulfamethazine in the indirect ELISA.The results demonstrate that the conjugate of SM2-HSA possess immunogenicity,which provides a basis for immunoassay of hapten-drug residue.
