北京、宁夏甜椒上分离的黄瓜花叶病毒CP基因序列分析及亚组鉴定
Sequence analysis of CP gene and identification of subgroup of cucumber mosaic virus from Beijing and Ningxia
【摘要】 从宁夏和北京地区甜椒上分离到3个黄瓜花叶病毒分离物(宁夏分离物为CMV-NX1,NX2;北京分离物为CMV-BJ)并保存在普通烟上。从叶片中提取该病毒RNA,经RT-PCR扩增到780bp的全长CP基因的cDNA,PCR产物纯化并测序。2个宁夏分离物(NX1,NX2)核甘酸序列同源性为100%,它们为同一株系。与已报道的部分CMV株系的CP基因序列相比较,表明3个分离物与CMV亚组I同源关系比亚组II更密切,因而3个分离物都属于黄瓜花叶病毒亚组I,通过DAS-ELISA进一步验证它们都属于亚组I。
【Abstract】 Three CMV isolates from pepper in Ningxia and Beijing were indentified as cucumber mosaic virus(CMV) by serology method and maintained in Nicotiana tabacum. The total RNA were extracted from infected leaves. The 780 bp cDNA product of CP gene was amplified by RT-PCR. Nucleotide sequence analysis and comparision of the coat protein (CP) gene of the three isolates were carried out after the 780 bp cDNA product was purified and sequenced. It showed that homology of the two Ningxia isolates were 100% at nucleotide level, which means the two isolates were of the same strain. The CMV-NX and CMV-BJ were found to be more closely related with CMV subgroup I, which were also confirmed by DAS-ELISA.
