颜面皮肤癌中NOS和VEGF的表达及其临床病理意义

【摘要】 目的 研究血管内皮生长因子(VEGF)、诱导型一氧化氮合酶(iNOS)和内皮型一氧化氮合酶(eNOS)在颜面皮肤癌中表达的相关性;探讨三者与颜面皮肤癌血管生成和临床病理特征的关系;研究一氧化氮(NO)和 VEGF的相互作用及 NO在 VEGF促肿瘤生长中的作用机制。方法 应用免疫组织化学方法检测47例颜面皮肤癌手术切除标本 VEGF、iNOS和 eNOS的表达,Ⅷ因子相关抗原(FⅧRAg)血管内皮细胞特异性染色计数肿瘤微血管密度(MVD)。结果 (1)32例颜面皮肤癌组织表达VEGF,30例表达iNOS,35例表达eNOS;(2)VEGF与iNOS的表达呈正相关,与eNOS的表达无相关;(3)VEGF、iNOS的表达与颜面皮肤癌MVD呈正相关, eNOS的表达与颜面皮肤癌MVD的差异无显著性意义;(4)VEGF的表达与颜面皮肤癌淋巴结转移和肿瘤分化程度及肿瘤浸润深度呈正相关;与临床分期无关。iNOS表达与颜面皮肤癌的分化程度及临床分期和有无淋巴结转移及肿瘤浸润深度正相关;eNOS表达与颜面皮肤癌临床分期和分化程度及有无淋巴结转移及肿瘤浸润深度均无关。结论 (1)iNOS对 VEGF的生成和发挥作用的过程有重要影响;(2)MVD随着VEGF和iNOS表达的增强而增加,说明两者对颜面皮肤癌血管生成具有促进作用。

【Abstract】 Objective To study the relation of protein expression of inducible nitric oxide synthase(iNOS), endothelial nitric oxide synthase(eNOS) and vascular endothelial growth factor (VEGF)in facial skin carcinoma and its relation to clinical pathology and angiogenesis.Methods Facial skin carcinoma tissue specimens were obtained from 47 patients undergoing operation. The expression of iNOS, eNOS and VEGF were studied by immunohistochemical technique. Microvessel density(MVD) was counted by endothelial cell immunostained by anti-FⅧRAg antibodies.Results (1)Positive VEGF and iNOS and eNOS immunostaining were detected in 32(68.09%),30(63.83%)and 35(74.47%)patients with facial skin carcinoma respectively;(2)Expression of VEGF had a significant positive relation to iNOS. but not to eNOS;(3)Cancer invasion and cancer MVD were positively correlated with expression of VEGF and iNOS,but not of eNOS;(4)In the clinicopathologic factors, lymph node metastasis and differentiated degree and invasion were significant correlated with VEGF expression,but there was no significantly correlation between the clinicopathologic stage and VEGF positive expression. Expression of iNOS was significantly correlated with differentiated degree,clinicopathologic stage,lymph node metastasis and invasion. There were no significant correlation between the clinicopathologic stage,differentiated degree,lymph node metastasis,invasion and eNOS positive expression.Conclusion iNOS,rather than eNOS play a positive role in facial skin carcinoma development.iNOS and VEGF have a correlation in their favoring angiogenesis of facial skin carcinoma.
