大鼠脑积水模型的建立

【摘要】 目的建立一种成年大鼠脑积水动物模型。方法雄性Wistar大鼠60只,随机分为实验组和对照组,其中实验组48只,再随机平均分为A、B、C、D四个亚组,应用显微外科技术向枕大池内注入25%白陶土混悬液0.1 mL,分别在白陶土注射后第1、2、4、6周行MRI检测,鉴定脑积水是否形成,并测定第三脑室层面侧脑室大小。对照组12只用同样方法向枕大池内注入生理盐水0.1 mL,2周后行MRI检测。并行病理切片检查。结果实验组有34只大鼠成功诱发脑积水,且脑室随着时间的延长而逐渐扩张。结论白陶土注射法可以制作确切的大鼠脑积水模型,特别适用于急慢性脑积水的研究。

【Abstract】 Objective To establish a sort of adult rat model of hydrocephalus. Methods 60 adult male Wistar rats were randomly divided into experimental group and control group. In the former group, 48 rats were randomly separated into 4 subgroups evenly, namely, group A, B, C and D, and then were used to construct animal models of hydrocephalus by injecting 0.1 mL volume of 25% kaolin solution, with the aid of a surgical microscope, into the cisterna magna. MRI was performed 1, 2, 4 and 6 weeks after kaolin injection respectively to obtain T2-weighted images of brain. The 12 controls were treated in the same way except for injection of saline and performance of MRI after 2 weeks. Results Hydrocephalus was induced in 34 rats whose ventricular system expanded as time goes on. Conclusion Injection of kaolin solution to establish a rat model of hydrocephalus is reliable, particularly suitable for the study of the pathological changes caused by acute and chronic hydrocephalus.
