柯萨奇病毒A组16型中国分离株(Cox.A16 SHZH00-1)全基因组序列测定及分析
Complete Genome Analysis of Coxsackievirus A16 SHZH00-1 Strain Isolated from the Mainland of China
【摘要】 对分离自2000年中国深圳地区手足口病患儿粪便标本的柯萨奇病毒A组16型(CoxsackievirusA16,Cox.A16)病毒SHZH00-1株进行全基因组序列测定及分析后发现,Cox.A16SHZH00-1的基因组(未包括多聚腺苷酸尾)长度为7410bp,其中5′端非编码区(5′UTR)长为745bp,病毒基因组编码区全长6582个核苷酸,编码一个含2193个氨基酸残基的多聚蛋白,3′端非编码区长为83bp。Cox.A16SHZH00-1基因组的结构与Cox.A16亚洲地方株Tainan-5079-98(AF177911)十分相近,整个基因组的核苷酸同源性为97.5%,氨基酸同源性为98.8%;而与Cox.A16国际标准株G10(U05876)差别较大,两者核苷酸同源性仅为79.1%,氨基酸同源性为94.5%。Cox.A16SHZH00-1与两株肠道病毒71型SHZH03(AY465356)和BrCr(U22521)比较,核苷酸同源性均不超过80%。
【Abstract】 Coxsackievirus A16(Cox.A16) is closely related to human enterovirus 71 and is often associated with outbreaks of hand-foot-and-mouth disease(HFMD).Cox.A16 infection is not always a benign infection, it can cause serious myocarditis and fatal shock.This study analyzed the complete nucleotide sequence of Cox.A16 SHZH00-1 strain isolated from Shenzhen 2000 in China. The results showed that the full genomic length of Cox.A16 SHZH00-1 is 7 410bp and the ORF is 6 579bp.The genome composition and construction are consistent with enterovirus. The nucleotide and amino acid sequences of Cox.A16 SHZH00-1, when compared with those of Cox.A16 Tainan-5079-98,G10, EV71 SHZH03 and BrCr strains, revealed that Cox.A16 SHZH00-1 is close to Tainan-5079-98,the nucleotide and amino acid identities are 97.5% and 98.8% respectively.Cox.A16 SHZH00-1 shares lower homology with G10,the nucleotide and amino acid identities are 79.1% and 94.5% respectively.Comparing Cox.A16 SHZH01 with EV71 SHZH03 and BrCr,the nucleotide identities are both less than 80%.
