GalNAcT和CK19mRNA表达与乳腺癌骨髓微转移的相关性

【摘要】 目的:研究乳腺癌患者骨髓中的β1,4N乙酰半乳糖胺转移酶(GalNAcT)mRNA和细胞角蛋白19(CK19)mRNA表达情况。方法:选取初治乳腺癌患者40例,在化疗前抽取骨髓,利用巢式逆转录聚合酶链反应(RTPCR)方法检测骨髓中GalNAcT和CK19的基因表达;另选择血液系统良性疾病患者10例和5名正常人作为对照组。结果:40例乳腺癌患者GalNAcTmRNA和CK19mRNA的阳性率分别为42.5%(17/40)和27.5%(11/40);10例血液系统良性疾病患者和5名正常人骨髓GalNAcTmRNA、CK19mRNA均不表达。两者表达与患者的TNM分期差异有显著性(P<0.05)。而与患者的年龄、肿瘤大小、腋窝淋巴结状态、病理类型以及激素状况等差异均无显著性(P>0.05)。结论:GalNAcTmRNA和CK19mRNA可作为标志物检测乳腺癌患者骨髓微转移,联合检测可增加骨髓微转移检测的敏感性。

【Abstract】 Objective:To study the relation of the expression of β1,4-N-acetylgalactosaminyl-transferase(GalNAcT) and cytokeration 19(CK19) mRNA and in breast cancer with micrometastases in bone marrow.Methods:RT-PCR was used to detect the expressions of GalNAcT and CK19 mRNA in the bone marrow from 40 patients with breast cancer.The bone marrow from 10 patients with benign blood diseases and the blood from 5 normal healthy volunteers served as controls.Results:The positive rates of GalNAcT and CK19 mRNA expressions in the bone marrow of the patients with breast cancer were 42.5%(17/40) and 27.5%(11/40) respectively.No expressions of GalNAcT mRNA and CK19 mRNA were found in the bone marrow of the 10 patients with benign diseases and the 5 healthy volunteers.The GalNAcT and CK19 mRNA expressions were correlated with TNM stages.There was no significant difference between the micrometastasis and other clinical parameters.Conclusions:The GalNAcT and CK19 mRNA expressions may be potential tumor markers for the detection of micrometastasis in the bone marrow of patients with breast cancer.
