高效液相色谱法测定犬血清中盐酸伪麻黄碱
Determination of pseudoephedrine hydrochloride concentration in dog serum by reversed phase high performance liquid chromatography
【摘要】 目的建立测定犬血清中盐酸伪麻黄碱的方法。方法采用高效液相色谱法,血清样品用乙醚提取,以ODSC18为固定相,甲醇∶乙腈∶磷酸盐缓冲液(pH3.5)=30∶10∶60为流动相,UV210nm紫外检测。结果在0.031252.0mg.L-1浓度范围线性良好,最小检测浓度为0.03125mg.L-1,最低检测限为0.625ng。其日内差(n=5)及日间差(n=5)RSD分别小于7.5%和7.6%,相对回收率(n=5)大于91.0%。结论方法简便灵敏,为盐酸伪麻黄碱的药代动力学研究提供了检测方法。
【Abstract】 AimTo establish a method for determination of pseudoephedrine hydrochloride in dog serum.MethodsPseudoephedrine was extracted with ethyl ether,and then measured by RP-HPLC using a ODS C18coWlumn asstationary phase and methanol-acetonitrile-phosphate buffer solution(pH 3.5)(301060)as mobile phase with ultraviolet Absorption at 210nm.ResultsThe method proved to be linear in the range of 0.031252.0 mgL-1and the lower limit of detection of pseudoephedrine hydrochloride in serum was 0.03125 mgL-1.The RSD was under 7.5%(n=5)and 7.6%(n=5)for inter-assay and intra-assay respectively,and the relative recovery rate was superior to 91.0%.ConclusionThis method is sensitive,specific,simple and validated for use in pharmacokinetic study.
