PCR-STR分型技术用于亲权鉴定疑难案1例调查

【摘要】 目的:对一例经血清学检测难以判断亲子关系的案例应用STR位点多态性进行亲权鉴定。方法:从冻存的当事人的样本中提取基因组DNA,体外扩增D19S253等13个STR位点DNA片段,扩增产物用聚丙烯酰胺凝胶电泳进行分离,银染显带。根据各STR位点基因型的检测结果判定亲子关系。结果:受检男孩在D1S1656、D8S1132、D8S1179、D21S11、FGA等5个位点与可疑父不符合遗传学规律,以及在D1S1656、D3S1358、D8S1132、D8S1179、vWA等5个位点与“亲生母亲”不符合遗传学规律,进而排除亲权关系。结论:STR位点具有高度的遗传多态性,聚合酶链反应检测STR位点基因型能够有效地开展亲权鉴定。

【Abstract】 Objective A case with serologically ambiguous typing results was typed again by PCR in STR loci for paternity testing. Methods The DNA extrated from frozen blood samples of the case was applied for PCR-based typing. PCR amplified products of 13 STR loci were analyzed by using the same PAG and silver staining. Results At least 5 STR loci incompatibilities between alleged father and the boy were found, and another 5 STR loci incompatibilities between"mother" and the boy were found. The results reach paternity exclusion standard. Conclusion The STR loci is highly polymorphic and suitable for paternity testing.
