电化学诱导铜(II)-磺基水杨酸配合物对DNA的切割
ELECTROCHEMICAL CLEAVAGE OF DNA IN THE PRESENCE OF COPPER-SULFOSALICYLIC ACID COMPLEX
【摘要】 研究了在铜(Ⅱ)-磺基水杨酸配合物存在情况下的电化学诱导DNA的切割.铜(Ⅱ)-磺基水杨酸配合物在控制的-0.4V电位下,Cu(ssal)2+2作为媒介通过电化学反应产生活性氧自由基O2·,氧自由基可以切割DNA.被切割的DNA片断使用高效液相色谱可进行分离.实验结果表明,电化学诱导铜(Ⅱ)-磺基水杨酸配合物对DNA的切割方法简单,切割效果好.
【Abstract】 Electrochemical cleavage of DNA in the presence of copper-sulfosalicylic acid (Cu(ssal)2+2) complex was studied. Cu(ssal)2+2 complex mediate generation of reactive oxygen species from O2 by the redox reaction in negative potential, these radicals are capable of damaging DNA. The cleaved DNA fragments were separated by high performance liquid chromatography (HPLC). The experimental results indicated that the method for electrochemical cleavage of DNA by Cu(ssal)2+2 complex was simple and efficient.
