余甘子减少高脂血症对兔动脉壁损伤作用的实验研究

【摘要】 目的探讨余甘子能否减少高脂血症对兔动脉壁的损伤及机制。方法将24只新西兰雄性白兔随机分为对照组、余甘子组(4g·kg-1·d-1)和高胆固醇血症模型组,每组8只。酶法检测3组血脂含量,8周后处死,组织匀浆测定主动脉和肝脏脂质含量,图像分析法测定主动脉内膜粥样硬化斑块面积和内膜面积与中膜面积,光镜观察动脉内膜病变。结果实验8周后余甘子组与高脂组相比,(1)能显著抑制血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)水平的上升,增加血清高密度脂蛋白胆固醇(HDL-C);(2)降低主动脉和肝脏脂质含量;(3)主动脉斑块面积、内膜厚度与中膜厚度比值明显降低。结论余甘子可能通过调整家兔脂质代谢,防止兔实验性粥样斑块的形成。

【Abstract】 Objective To study the protecting effect of Phyllanthus embica (PE) on arterial wall in hyperlipemia rabbit model. Methods Twenty-four male New Zealand rabbits were randomized into three groups:the control group was fed with com- mon diet for 8 weeks;hypercholesterolemia group and PE groups were fed with 0.5 % cholesterol for 8 weeks.In PE group 4 g/(kg·d) PE was added.At the end of the 8 weeks,rabbits were killed.The serum levels of triglyceride (TG),total cholesterol (TC),low density hpoprotein cholesterol(LDL-C) and high density lipoprotein cholesterol(HDL-C) were mea- sured before experiment,4th and 8th week of experiment by enzyme dynamics method.The lipid of aorta and liver tissue were also evaluated at the 8th week.Light microscopy was adopted to assess the degree of pathologic damage of arterial wall.Results As compared with the hypercholesterolemia group:(1) PE could reduce the levels of serum TC and LDL- C (P<0.05),but elevate HDL-C level significantly (P<0.05),the contents of lipid of aorta and liver tissue were de- creased markedly (P<0.05).(2) PE could also reduce the extent of atherosclerotic lesion and the aorta plaque areas significantly(P<0.05).Conclusion PE has the effect of inhibiting development of experimental atherosclerosis,this action may be attributed to its function on regulating lipid metabolism.
