工业化生产EPA和DHA藻株的选育

【摘要】 为筛选出高产EPA和DHA藻株 ,对 8种海洋微藻 (硅藻门 3株 ,绿藻门 2株 ,金藻门 2株 ,红藻门 1株 )进行培养 ,测定了它们的最佳吸收波长和生长曲线。藻体在对数期末收获 ,经过提取脂肪 ,进行皂化、酯化处理后用气相色谱测定EPA和DHA的含量。结果表明 ,EPA高产藻株为新月菱形藻 ,产量为 16mg/L ,占粗脂肪重的 2 6 % ,占细胞干重的 3.3% ;等鞭金藻为DHA高产株 ,DHA产量为 3.2mg/L ,占粗脂肪重的 9.1% ,占细胞干重的 5 .2 %。

【Abstract】 Eight algal strains were cultured for the selection of algal strains with high production of EPA and DHA and the optimum absorb wavelength and growth curve were determined.They were harvested at the end of exponental period.Then lipids components were extracted.The extracted lipids were transmethylated,and the methyl esters were analyzed by gas chromatography to determine the contents of EPA and DHA.The results show that the most EPA production is Nitzschia closterium,with the content of 16 mg/L,representing 26% of cellular total fatty acid content,and 3.3% of celluar dry weight(DW).The most DHA production is Chrysophyceae isochrysis galbana,and its content is 3.2 mg/L,representing 9.1% of total fatty acid,and 5.2% of DW.
