粉防己碱治疗大鼠移植心脏慢性排斥的实验研究
Experimental Study of Tetrandrine on Chronic Rejection in Heterotopic Heart Transplantation of Rat
【摘要】 目的 应用大鼠异位心脏移植模型研究粉防己碱 (TET)对慢性排斥反应 (CR)的抑制作用。方法 异品系颈部心脏移植 (SD大鼠移植于Wistar大鼠 )受体鼠每日给予环孢霉素A (CsA)灌胃 (1.5mg/kg)以诱导CR ;异品系大鼠分两组 :2 5mg/kgTET组大鼠同时每日给予 2 5mg/kgTET灌胃 ,5 0mg/kgTET组同时给予 5 0mg/kgTET灌胃 ;Wistar大鼠间的心脏移植 (同系对照组 )不用任何药物治疗。 12周后取移植心脏做组织学检查 ,以HPIAS— 10 0 0图像分析仪测量动脉管壁与血管直径的比值。结果 对照组移植心脏可见明显的纤维化 ,大量单核细胞浸润 ,动脉硬化明显 ,动脉管壁与血管直径的平均比值明显高于同系对照 (0 .64± 0 .2 0vs0 .2 3± 0 .0 7,P <0 .0 1)和TET 5 0mg /kg组 (0 .64± 0 .2 0vs0 .3 3±0 .11,P <0 .0 5 ) ,TET 2 5mg/kg组的动脉管壁与血管直径的平均比值为0 .40± 0 .13 ,高于同系对照组 (P <0 .0 5 ) ,但与异系对照组及TET 5 0mg治疗组比较无统计学意义。 结论 TET具有明显的抗移植物CR作用 ,其主要作用机制有待进一步研究。
【Abstract】 Objective Using rat heterotopic cardiac allografts transplantation model, the efficacy of tetrandrine (TET) in treatment of chronic rejection was investigated.Methods Cervical heterotopic heart transplantation was performed from SD to Wistar rats (allograft), and Wistar to Wistar rats (isograft controls). All allograft recipients were treated with CsAat a dose of 1.5mg/kg body weight. 25mg TET group rats were administered with TET at a dose of 25mg/kg body weight for 12 wk continuously, and 50mg TET group rats at a dose of 50 mg/kg. Isograft control group rats were not treated with anydrug. Histological changes were monitored 12 wk after operation, and the mean ratio of depth of vessel wall and vessel diameter was calculated with computer imaging analysis system (HPIAS-1000).Results In allografts controls, there were massive fibrosis, monocytes infiltration, and arteriosclerosis compared with TET-treated groups and isograft control group; the mean ratio of depth of vessel wall and vessel diameter was 0.64±0.20, much higher than that in isografts (0.23±0.07,P<0.01) and 50 mg TET group (0.33±0.11,P<0.05), the mean ratio in 25mg TET group was 0.40±0.13, higher than that in isograft group (P<0.05), but there was no statistic difference compared with isograft or 50mg TET group.Conclusion TET may attenuate the development of chronic rejection, and the mechanisms may be related with TET possessing effects against ischemia reperfusion injury, and inhibiting the expression of cytokines.
