谷胱甘肽合中药对帕金森病动物模型抗氧化指标及细胞形态学的影响
The Effect of Glutathione Combined with Traditional Chinese Medicine on Antioxidation Index and Cell Morphology of PD Model Rats
【摘要】 目的 :观察还原型谷胱甘肽 (GSH)合中药抗震止痉胶囊对帕金森病 (PD )大鼠模型的异常形为、黑质抗氧化系统及细胞形态学的影响 ,并探讨其作用机制。方法 :应用 6-羟基多巴胺立体定向注射制作PD大鼠模型。将2 4只大鼠随机分为 3组 (每组 8只 ) :模型组、治疗组 (GSH合抗震止痉胶囊组 )、假手术组 ,分别给予相应处理 ,共 45d ,给药前后均进行行为学测试 ,给药结束后每组随机选取 2只大鼠 ,分别行免疫组化和电镜观察 ,测定其余大鼠黑质区GSH -Px、MDA及活性氧水平。结果 :①治疗组可明显改善大鼠旋转行为 (P <0 .0 5 ) ;②治疗组能减轻黑质区氧化应激损伤 ;③免疫组化发现治疗组TH -IR神经元较模型组明显增多 (P <0 .0 0 1) ;④电镜下发现治疗组可部分延缓凋亡进程。结论 :GSH合抗震止痉胶囊能部分改善大鼠旋转行为 ,减轻PD模型大鼠黑质区氧化应激损伤 ,并对残存DA能神经元具有一定保护作用。
【Abstract】 Objective:To observe the effects of Glutathione Combined with Kang-zhen Zhijing Capsule on the abnormal behavior, nigral antioxidation system and cell morphology of PD model rat and study its mechanism. Methods:The model rats for PD were established through stereotoxic microinjection of 6-hydroxydopamine (6-OHDA). The 24 model rats after behavior test were randomly divided into 3 groups: Model group(MG), Glutathione Combined with Kang-zhen Zhijing Capsule group (GKG) and sham-operated group (SG). Every group was given relevant treatment for 45 days. All rats behavior were tested before and after giving medicine. 2 rats randomly chosen from every group were observed under immunohistochemistry and electron-microscope. The activity of GSH-Px, content of MDA and ROS of nigra of all other rats were assayed. Results:①GSH Combined with Kang-zhen Zhijing Capsule had obvious effect on its rotation behavior; ②Glutathione Combined with Kang-zhen Zhijing Capsule could decrease the nigral oxidative stress of PD model rats; ③The TH-IR neurons of damaged side of GG increased compared with MG (all P<0.001); ④GSH could partly postpone the course of apoptosis. Conclusion:GSH Combined with Kang-zhen Zhijing Capsule could ameliorate partially PD model rats rotation behavior, decrease the nigral oxidative stress of PD model rats, and protect surviving dopaminergic neurons.
