川芎内酯A预处理对大鼠离体心脏缺血再灌注损伤的保护作用

【摘要】 目的 :观察川芎内酯A预处理对离体心脏缺血再灌注损伤的保护作用。方法 :采用Langendorff离体心脏恒压灌流方法 ,以 0 .5ml/min的灌流速度、浓度分别为 0 .0 12 5mg/ml、0 .0 2 5mg/ml和 0 .0 5mg/ml的川芎内酯A ,与K H灌流液同时灌流心脏10min ,然后全心停灌 3 0min ,复灌 60min。结果 :川芎内酯A预处理可提高心脏缺血再灌期的冠脉流量和心肌收缩力 ,降低室颤和室速的发生率 ,使心肌匀浆的乳酸脱氢酶、丙二醛含量降低 ,超氧化物歧化酶活性增强。结论 :川芎内酯A预处理对离体心脏缺血再灌注所致损伤可能具有保护作用。

【Abstract】 The aim of this study was to determine whether pharmacologic preconditioning, without a short episode of myocardial ischemia, could improve myocardial function after a prolonged period of ischemia. Isolated rat hearts were perfused with 0.0125 mg/ml, 0.025mg/ml, 0.05mg/ml for 10min followed by a 10min washout period before the induction of 30min normothermic global ischemia and 60min of reperfusion. Hearts preconditioned with chuanxiong pathalide produced a reduction in the incidence of reperfusion induced ventricular fibrillation (VF) and ventricular tachycardia (VT). Preconditioning of the hearts with highest dose of chuanxiong pathalide (0.05mg/ml), after 30min of ischemia, reduced the incidence of reperfusion induced VF and VT from their nonpreconditioned control value of 100% to 37.5% (P<0.05),and delayed the onset time of VF and VT from nonpreconditioned control average value of 1.82±0.74min to 39.21±10.21min. After 30min of ischemia, the recovery of myocardial function was significantly improved in chuan xiong pathalide preconditioned group. Thus, 0.0125 mg/ml, 0.025mg/ml and 0.05mg/ml of chuanxiong pathalide shortened the duration of VF and VT from their nonpreconditioned control value of 23.51±3.99min to 7.50±1.61min, 1.64±0.67min, 1.06±0.70min,respectively, 0.05mg/ml of chuanxiong pathalide increased aortic flow and content of SOD from its nonpreconditioned control value of 59.6±18.7 U/mgprot to 92.3±19.0 U/mgprot with decreased release of LDH and MDA from their nonpreconditioned control value of 2378.0±196.3 U/gprot and 12.1±1.3 nmol/mgprot to 1669.4±192.5 U/gprot and 6.9±0.8 nmol/mgprot, respectively. Our results show that chuanxiong pathalide preconditioning can prevent postischmic abnormalities in reperfusion arrhythmias, aortic flow and myocardium metabolism.
