金雀异黄素抑制ox-LDL诱导的人脐静脉平滑肌细胞MCP-1表达的信号途径初探

【摘要】 目的 研究金雀异黄素抑制氧化型低密度脂蛋白诱导的人脐静脉平滑肌细胞单核细胞趋化蛋白 1mRNA和蛋白表达的作用机制。方法 采用逆转录聚合酶链反应技术和酶联免疫吸附试验测定细胞中单核细胞趋化蛋白 1mRNA的表达量及蛋白的含量。观察雌激素受体拮抗剂他莫昔芬对金雀异黄素抑制单核细胞趋化蛋白 1表达作用的影响。结果 他莫昔芬不能阻断金雀异黄素抑制单核细胞趋化蛋白 1mRNA和蛋白表达的作用(P >0 .0 5 )。结论 金雀异黄素下调氧化型低密度脂蛋白诱导的人脐静脉平滑肌细胞单核细胞趋化蛋白 1的表达可能主要是通过非雌激素受体途径所介导
【Abstract】 Objective To study the mechanism of genistein inhibiting the expression of monocyte chemotactic protein-1 (MCP-1) mRNA and protein induced by oxidized low density lipoprotein (ox-LDL) in human umbilical vein smooth muscle cells (hUVSMC). Methods Reverse transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA) were adopted to measure expression of MCP-1 mRNA and protein. The effect of antiestrogen tamoxifen on genistein inhibiting the MCP-1 expression was observed. Results Tamoxifen could not reverse the genistein inhibition of MCP-1 expression (P>0.05). Conclusions It is possible that mainly through estrogen receptor independent pathway, genistein could down-regulate the expression of MCP-1 in hUVSMC stimulated with ox-LDL.
