HPLC法测定瑞格列奈及片剂中的R-异构体
Determination of R(-)-Isomer in Repaglinide and Its Tablets by HPLC
【摘要】 目的建立HPLC法测定瑞格列奈及片剂中R异构体。方法用α酸性糖蛋白键合硅胶为固定相,0.02mol·L-1的磷酸盐缓冲液(pH7.0)乙腈(90∶10)为流动相,流速0.5ml·min-1,检测波长240nm。结果瑞格列奈与R异构体分离良好,最低检出量为1ng,R异构体的回收率为99.0%。结论方法快速简便,准确,可用于瑞格列奈及片剂中R异构体的测定。
【Abstract】 Objective: To establish a HPLC method for determination of R ( ) isomer in repaglinide and its tablets.Method: A Chiral AGP column was used with 0.02 mol·L -1 buffer of potassium phosphate(pH 7.0 ) acetonitrile as mobile phase. The flow rate was 0.5 ml·min -1 and the detecting wavelength was at 240 nm.Result: The detecting limit of nateglinide was 1ng and the recovery of R ( ) isomer was 99.0% .Conclusion: This method is rapid, simple, and can be used for determination of R ( ) isomer in repaglinide and its tablets.
