青藤碱的身体依赖性和精神依赖性实验研究
An Experimental Study on Physical and Psychological Dependences of Sinomenine
【摘要】 目的:观察青藤碱是否具有身体依赖性及精神依赖性,评价其安全性。方法:采用大、小鼠催促戒断模型、大鼠自然戒断模型及小鼠条件性位置偏爱模型检测青藤碱的药物依赖性。结果:在催促戒断试验中,受试的大、小鼠连续注射青藤碱10天后经纳洛酮催促,均未出现戒断症状及体重下降现象。在大鼠自然戒断试验中,青藤碱低(总给药剂量为1.05 g·kg-1)、高(总给药剂量为2.1 g·kg-1)两个剂量连续使用28天,停药后大鼠没有出现戒断反应及体重下降现象。在小鼠条件性位置偏爱试验中,青藤碱低、中、高三个剂量组分别连续用药6天后,小鼠在伴药盒的逗留时间均无明显延长,不形成条件性位置偏爱。结论:青藤碱不具有身体依赖性及精神依赖性,无成瘾性,使用该药安全性大。
【Abstract】 Objective: To investigate the physical and psychological dependences-the ability of producing addiction of sinomenine and evaluate its drug safety. Methods: Two naloxone-precipitated tests in morphine-dependent mice and rats, a spontaneous withdrawal test in rats and a conditioned place preference test in mice were used in this study to detect the dependences of sinomenine. Results: In the naloxone-precipitated tests, after injecting sinomenine for 10 days continuously, the mice and rats didn’t show any withdrawal symptom and the loss of body weight. In the spontaneous withdrawal test, two dosages of sinomenine (total dosage, respectively of 1.05 g·kg-1 and 2.1 g·kg-1) were continuously given for 28 days. The withdrawal response and the loss of body weight were not found in sinomenine-treated rats after drug withdrawal. In the conditioned place preference test, the time of staying in the drug-pair compartment wasn’t significantly extended and place preference effect wasn’t developed in three dosages of sinomenine-treated mice. Conclusion: Sinomenine doesn’t result in the physical or psychological dependence or any addiction in rats and mice in this study which suggests sinomenine be safe for use.
